The Smile Has Left Your Eyes bercerita mengenai dua orang yang ditakdirkan untuk bertemu dan terus berpapasan di kehidupan sehari-hari.
Seorang detektif Yoo Jin Kook (Park Sung Woong) terobsesi dalam memecahkan kasus pembunuhan seorang mahasiswi yang disamarkan sebagai bunuh diri. Ia bertemu seorang lelaki yang dicurigai olehnya terlibat dalam kasus pembunuhan tersebut.
Lelaki tersebut, Kim Moo Young (Seo In Gook) bertemu dengan adik perempuan detektif tersebut, Yoo Jin Kang (Jung So Min) dan akhirnya saling jatuh cinta. Yoo Jin Kook berusaha menjauhkan Yoo Jin Kang dan Kim Moo Young karena kecurigaannya.
